在加速剂作用下 ,间接胆红素内氢键被破坏,使非水溶性的间接胆红素与直接胆红素在酸性条件下与偶氮试剂反应生成偶氮胆红素,生成的偶氮胆红素在565 nm处有最大吸收,测定吸光度变化可求出血清中总胆红素含量 。
